We are installing bamboo floors in our bedroom and need to know if it's necessary to make sure the over spray of paint from the builders needs to be removed before applying the glue. It's very thin and no thick sections or drips just a light powder like over spray.

You are Probably ok with the over spray, just sweep well, removing the dust before applying the glue.
